IHRC, Inc. provides scientific, information management, management consulting, bioinformatics, and administrative program support to the Centers for Disease Control and Prevention (CDC) and other Federal and private sector clients.  The successful candidate will be a full-time employee of IHRC, Inc., and will be assigned to the Laboratory of Infection Control, Office of Science and Engineering Laboratories (OSEL), Division of Biology, Chemistry and Material Sciences (DBCMS), at the Food and Drug Administration (FDA), 10903 New Hampshire Avenue, Silver Spring, MD 20993, to design, develop and optimize sensitive, specific, and robust, in vitro diagnostic assays.

SCOPE OF WORK

The Visiting Scientist will design, develop and optimize sensitive, specific, and robust, in vitro diagnostic assays that will clearly differentiate between Zika Virus (ZV) and the closely related West Nile Virus (WNV) and Dengue Virus (DV), and can be used in BSL-2 to identify ZV antibodies in infected patients.

MAJOR DUTIES AND RESPONSIBILITIES

  • Develop a Zika sensitive mouse model and Zika reagents for nucleic acid assays and immunoassays

    • Develop a Zika sensitive rodent model
    • Generate Zika specific plasmids and proteins and conduct the immunization of mice and rabbits with Zika proteins
  • Develop Zika specific cell mediated immunoassays
    • Develop a Zika ELISpot assay, conduct bioinformatic analysis of Zika peptides binding with Class 1 MHC of Rhesus Macaque MHC (Mamu) and perform Zika peptide stabilization studies
    • Develop a Zika mimotope library for proteins and identify the cytokine expression in T cells following Zika immunization using a mimotope library
  • Develop Zika Specific humoral immunoassays
    • Develop Zika ELISA and Luminex-based assays, identify immunodominant antibody epitopes using a slide based peptide library, construct Zika plasmids for virus like particles (VLPs) and develop Zika based VLP neutralizing antibody assays using a mimotope library for proteins
  • Author detailed laboratory reports with plots, charts, and tables illustrating the performance of the Zika specific immunoassays
    • Collate, synthesize and interpret data collected and compile into scientific manuscripts suitable for publication

Qualifications:

MINIMUM QUALIFICATIONS

Education and Experience:

  • Ph.D. in immunology, biotechnology, biochemistry, microbiology, molecular biology or a related discipline with 5 years of experience performing research related to the requirements of the position

Required:

  • Demonstrated experience with research involving cellular and humoral immunity immunological assays, and small animal handling
  • Suitable training and publications demonstrating the ability to:
    • Identify/generate a Zika sensitive/transgenic rodent model, generate/amplify and purify plasmids and generate antibody responses in mice
    • Utilize bioinformatics software to identify potential immunodominant epitopes, establish flow based Class 1 MHC stability studies, establish a flow cytometric based cytotoxic T cell assay, generate and characterize a mimotope library, and generate tetramers to follow cell mediated responses in mice
    • Employ Luminex technology, identify humoral epitopes using peptides printed on slides, and generate virus like particles (VLPs) for flow based neutralizing assays
    • Write detailed monthly summaries of ongoing projects
  • Excellent writing and documentation skills, and ability to achieve goals with minimal supervision

Desirable :

  • Previous experience developing intracellular cytokine assays and multi-colored flow cytometry based immune assays to measure Zika envelope antigen- specific responses
  • Strong publication record in high impact scientific journals demonstrating sound theoretical and practical knowledge of immunology, cell biology and molecular biology
  • Expertise in developing assays related to antigen-specific antibody detection and quantification in serum and mucosal samples
  • Experience and expertise working with multiple animal models using a variety of techniques to study the cellular and humoral immune response
  • Experience and expertise in the authoring and authorization of Institutional Animal Care and Use Committee (IACUC) protocols

Language Skills:

  • The candidate must possess excellent oral and written communication skills in English
